氯化锑对斑马鱼胚胎发育的影响

【摘要】目的研究氯化锑(SbCl_3)对斑马鱼胚胎发育的影响。方法将单细胞期的斑马鱼胚胎暴露于400、600和800μmol/L的SbCl_3 24 h后观察胚胎死亡率。选取非致死剂量(400μmol/L)的SbCl_3暴露24、48和72 h后观察斑马鱼的胚胎形态变化。采用实时定量PCR(qRT-PCR)检测胚胎凋亡基因表达,吖啶橙(AO)染色检测凋亡。采用qRT-PCR检测胚胎自噬相关基因表达水平,免疫印迹检测自噬相关蛋白质LC3、p62表达水平。结果 400μmol/L SbCl_3暴露24 h有增加胚胎死亡率的趋势,但差异未见统计学意义(P=0.103);而600和800μmol/L SbCl_3暴露24 h较对照组明显增加胚胎死亡率(P<0.01)。400μmol/L SbCl_3暴露24、48和72 h均未见明显影响胚胎形态。400μmol/L SbCl_3暴露72 h上调凋亡相关基因caspase9(P<0.05)、p21(P<0.01)和Bax(P<0.01)的表达;而caspase3、caspase8和p53基因mRNA表达水平未发生明显变化(P>0.05); AO染色未见明显凋亡。此外,400μmol/L SbCl_3暴露72 h上调自噬相关基因lc3b(P<0.05)、beclin1(P<0.01)和Atg12基因mRNA表达(P<0.01),但不影响Atg3的mRNA表达水平(P>0.05);同时,该暴露条件上调LC3-II(P<0.05),下调p62蛋白表达水平(P<0.01)。结论 SbCl_3浓度依赖地增加斑马鱼胚胎死亡率。400μmol/L SbCl_3暴露未对斑马鱼胚胎形态造成明显影响;但上调caspase9、p21、Bax的表达,诱导斑马鱼胚胎发生自噬。
