石斛兰“绿色星辰×出水芙蓉”杂交种子无菌萌发与组培快繁研究(英文)

【摘要】[目的]研究石斛兰"绿色星辰×出水芙蓉"杂交种子无菌萌发与组培快繁。[方法]以"绿色星辰"为母本,以"出水芙蓉"为父本进行杂交,对其杂交种子进行了离体培养和组培快繁研究。[结果]石斛兰杂交种"绿色星辰×出水芙蓉"的F1代种子在改良Knudson培养基上(附加1.0mg/L6-BA、1.0mg/LNAA和10%熟香蕉泥)可获得理想的无菌萌发效果,且原球茎在该培养基上也能较快增殖。以1/2MS最适合于该杂种石斛兰F1代无菌实生苗的组培快繁;8种供试培养基用于增殖培养的效果从优到劣依次为:1/2MS>MS>1/3MS>1/4MS≈N6>改良Knudson≈B5>H。NAA用于杂种石斛兰"绿色星辰×出水芙蓉"F1代无菌实生苗的生根壮苗效果要优于IBA;NAA的推荐浓度为2.0mg/L。[结论]为今后的新品种选育提供了育种材料。

Rainbow2compactum ] under aseptic condition and to explore the parameters fo r rap id p ropagation of F1 plantlets via tissue culture. [M ethod] Hybridization between Den. Burana Green Star(female parent) and Den. Rainbowcompactum (male parent) was perfo rmed and the in vitro culture and pro liferation of F1 hybrids were studied using eight different basic media including MS, 1 /2MS, 1 /3MS, 1 /4MS,B5 ,N6 ,modified Knudson and H. [ Result] Imp roved Knud2son medium appended w ith 1. 0 mg /L 62BA, 1. 0 mg /L NAA and 10% mature banana puree perfo rmed best in F 1 seed germ ination under aseptic condition, as well as the rapid propagation of p ro tocorm2like body. O f all the eightmedia tested, 1 /2MS is the medium mo st suitable for the in vitro rap id p ropagation of the F1 seedlings.
